Toxicity and Biosafety of Houttuynia cordata
Toxicological safety studies of Houttuynia cordata have shown low overall toxicity of the plant upon oral and external use. In a study conducted on laboratory animals (mice and rats), it was determined that the median lethal dose (LD₅₀) of the aqueous extract of Houttuynia cordata exceeds 10 g/kg body weight upon oral administration, which indicates extremely low acute toxicity. When used in the form of alcohol tinctures or ethanol extracts at doses up to 2 g/kg, no signs of hepatotoxicity, nephrotoxicity, or neurotoxicity were detected.
Skin tests also did not demonstrate sensitizing or phototoxic effects, provided recommended dosages are observed. Data on embryotoxicity and teratogenicity are absent, however, caution is required during pregnancy and lactation due to limited observations.

Pharmacodynamics — Houttuynia cordata
Houttuynia cordata is a plant with a complex biochemical composition that exerts a modulating effect on several human physiological systems, including the immune, inflammatory, skin, respiratory, gastrointestinal, and nervous systems. The basis of pharmacological activity is composed of flavonoids (quercetin, rutin, hyperoside), essential oils (methyl nonyl ketone, lauric acid), polysaccharides, as well as phenolic compounds with high bioreactivity.
One of the key effects of Houttuynia cordata is a pronounced anti-inflammatory action, realized through inhibition of the production of pro-inflammatory cytokines (including TNF-α, IL-6, IL-1β) and a decrease in the activity of the nuclear factor NF-κB. The plant affects MAPK and JNK signaling pathways, preventing the activation of transcription of inflammatory genes, as well as reducing the expression of COX-2 and iNOS enzymes involved in the development of the local inflammatory cascade. It has been established that extracts of Houttuynia cordata can suppress the synthesis of prostaglandins and nitric oxide in macrophages, which confirms a systemic and local anti-inflammatory effect.
At the level of the immune system, the plant exhibits immunotropic properties, promoting the activation of phagocytosis, enhancing the activity of NK cells, and stimulating the production of immunoglobulins. Polysaccharides of Houttuynia cordata are involved in the regulation of the T-cell response, increasing the proliferative activity of lymphocytes and stimulating the production of interleukins under conditions of immune suppression. The immunomodulatory effect is combined with pronounced antioxidant activity, based on the ability of flavonoids to neutralize free radicals and enhance the activity of endogenous antioxidant enzymes, including superoxide dismutase and catalase.
Among the local effects, the antimicrobial and fungistatic action of Houttuynia cordata deserves attention, due to disruption of the integrity of bacterial cell walls, inhibition of biofilms, and suppression of the growth of yeast and mold fungi. The essential oils of the plant disrupt the ion permeability of pathogen membranes, which has been confirmed in vitro on a number of strains of gram-positive and gram-negative bacteria. The lipophilic fractions of the plant penetrate the lipid layers of the epithelium and act as a local antiseptic, especially with external use.
From the gastrointestinal tract, a normalizing effect on motor and secretory functions has been noted, in particular through local anti-inflammatory and antioxidant effects on the mucous membrane. Some polysaccharide components have a prebiotic effect and influence the regulation of intestinal microbiota, maintaining a balance between obligate and facultative flora. This is accompanied by a decrease in the production of metabolites of bacterial inflammation and an improvement in the trophism of the mucous layer.
With respect to the nervous system, the pharmacodynamics of Houttuynia cordata are mediated by a mild sedative effect. Flavonoid compounds, including hyperoside and rutin, affect the activity of GABA receptors and reduce the level of neuronal excitability. The neuroprotective effect is also realized through an antioxidant mechanism, preventing damage to neuronal membranes by reactive oxygen species. Along with this, a stabilizing effect on cerebral vascular tone is noted, associated with modulation of intracellular calcium levels and a decrease in capillary permeability.
Thus, the pharmacodynamic profile of Houttuynia cordata includes systemic anti-inflammatory, immunomodulatory, antiseptic, antioxidant, neuroprotective, and mild sedative effects, realized at the level of both cellular and humoral components of physiological regulation. Targets include inflammatory mediators, pro-oxidant molecules, cell receptors, enzyme systems, and epithelial barrier structures.

Pharmacokinetics — Houttuynia cordata
The pharmacokinetic properties of Houttuynia cordata have not been fully characterized, however, generalized data on the main groups of active substances and dosage forms allow describing the proposed mechanisms of absorption, distribution, metabolism, and elimination. The main biologically active compounds of the plant are flavonoids, essential oils, terpenoids, phenolic acids, and polysaccharides. Their pharmacokinetic behavior varies depending on the form of administration and the method of preparation of the medicinal product.
With oral use (in the form of powder, dry extract, aqueous or alcohol tinctures), the absorption of polyphenolic compounds and flavonoids occurs mainly in the small intestine with the participation of passive diffusion and transporter-mediated transport. A significant part of glycoside flavonoids undergoes hydrolysis in the intestine with the participation of microflora, which affects the composition of absorbed metabolites. High molecular weight polysaccharides are poorly absorbed and exert their effect mainly in the intestinal lumen and on the mucous membrane. Some compounds, such as essential oils, are rapidly absorbed through the gastrointestinal mucosa and begin to circulate systemically, including hepatic metabolism.
With transdermal and external use (ointment, cream, oil infusion, serum), the absorption of active compounds depends on their lipophilicity. Essential oils and some flavonoids penetrate the epidermal barrier and reach the deeper layers of the skin. At this level, they can exert local anti-inflammatory and antimicrobial effects. Accumulation in skin tissues is possible with prolonged use, especially in areas with increased permeability. Entry into the systemic circulation with external use is considered limited, but is possible when applied to damaged skin or mucous membranes.
With intravaginal or rectal administration (suppositories), absorption occurs mainly through the rich vascular networks of the mucous membranes, bypassing the hepatic first pass. This increases the systemic bioavailability of terpenes and flavonoid aglycones, especially when applied against the background of mucosal inflammation. Metabolism occurs partially in the mucosal tissues, but the main biotransformation is carried out in the liver with the participation of phase I and II enzymes (oxidation, conjugation, glucuronidation).
The liver is the main organ of metabolic transformation of flavonoids, terpenes, and phenolic compounds of Houttuynia cordata. Conjugated metabolites are excreted primarily with bile and urine. Some volatile components and their metabolites may be partially excreted through the lungs and skin. Polysaccharides and unabsorbed fragments of the plant cell wall are excreted in feces unchanged. The level and rate of excretion are influenced by the state of the intestinal microbiota and the functional activity of the liver and kidneys.
Thus, the pharmacokinetics of Houttuynia cordata is a multi-level process involving mucous membranes, the liver, intestinal microflora, the skin barrier, and the renal-biliary excretion system. The main routes of administration — oral, transdermal, and mucosal — provide both local and systemic distribution of active compounds, provided the preparation technology is observed.

Mechanisms of Action and Scientific Rationale — Houttuynia cordata
The pharmacological activity of Houttuynia cordata is due to the combined action of polyphenolic compounds, terpenes, polysaccharides, and volatile essential components, most of which exhibit a regulatory effect on key signaling cascades of inflammation, immune response, and cellular defense. One of the central mechanisms of action of the plant extracts is considered to be the inhibition of the nuclear transcription factor NF-κB, which leads to a decrease in the expression of pro-inflammatory cytokines (including TNF-α, IL-1β, IL-6) and nitric oxide synthase (iNOS). It has been established that fractions of H. cordata inhibit the activation of the MAPK pathway, including p38 and JNK, blocking the intracellular transmission of signals associated with inflammation and cell proliferation.
In parallel, an effect on the enzyme systems of cyclooxygenase (COX-2), lipoxygenase (5-LOX), as well as inhibition of the production of prostaglandins and leukotrienes is observed, which confirms the anti-inflammatory and analgesic action at the systemic and tissue levels. The essential oils of the plant demonstrate activity against gram-positive and gram-negative microorganisms by disrupting the integrity of bacterial membranes, inhibiting the activity of bacterial enzymes, and inhibiting the synthesis of nucleic acids. An inhibitory effect on the formation of biofilms and the expression of antibiotic resistance genes has also been established, which makes H. cordata a promising component of antimicrobial compositions.
On the immune system, H. cordata has a complex effect. In in vitro and in vivo models, extracts enhanced the phagocytic activity of macrophages, stimulated the production of Th1-type cytokines, including interferon-gamma (IFN-γ), and activated NK cells. This indicates an immunomodulatory effect with a predominance of cellular response. Plant polysaccharides activate Toll-like receptors (TLR2 and TLR4) on the surface of immune cells, initiating signaling cascades involving the NF-κB transcription complex, which contributes to the restoration of disturbed immune balance. The effect on the JAK/STAT system may explain the observed antiproliferative action on inflammatory-activated cells.
The antioxidant activity of Houttuynia cordata is realized through direct neutralizing effects on reactive oxygen species (ROS) and induced increases in the activity of antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx). Due to this, the plant is able to reduce oxidative damage to lipid membranes, proteins, and DNA. Flavonoids, including rutin and hyperoside, interact with metal cations and free radicals, stabilizing them and preventing cascades of oxidative stress.
In addition, modulating effects on signaling systems regulating apoptosis have been identified. In particular, components of H. cordata increase the expression of p53 and Bax proteins, while simultaneously suppressing Bcl-2 expression, which leads to the activation of the mitochondrial pathway of apoptosis. This may be associated with antiproliferative activity and participation in the control of the cell cycle.
Some studies show that plant extracts interact with enzymes that regulate neurotransmitter levels, including inhibition of acetylcholinesterase (AChE) activity and potential binding to GABA and 5-HT receptors, which partially explains the sedative and neurotropic action with systemic administration.

Synergy — Houttuynia cordata
According to modern pharmacological studies, Houttuynia cordata exhibits pronounced synergistic activity in combination with a number of plant and biologically active substances, which allows potentiating its physiological effects at the molecular and cellular levels. The most reliable data have been obtained on interactions with flavonoids, polysaccharides, terpenoid compounds, and bacteriostatic agents of natural origin. In particular, in vitro studies have confirmed the potentiating effect of the combination of H. cordata extract with Scutellaria baicalensis extract through additive inhibition of the expression of pro-inflammatory cytokines (IL-6, TNF-α) and suppression of NF-κB activation. The combination affects similar signaling cascades, enhancing the anti-inflammatory and antioxidant action at the cellular level.
Combinations of Houttuynia cordata with extracts of Glycyrrhiza uralensis, Isatis tinctoria, and Lonicera japonica demonstrate additive and modulating interaction with respect to antimicrobial activity. Such combinations are capable of inhibiting the growth of pathogenic bacteria and viruses more effectively than the isolated use of individual components. The enhancement of action is achieved through multiple inhibition of bacterial enzymes, disruption of the lipid balance of the microbial membrane, and suppression of the transcription of resistance genes. In addition, such complexes affect the secretion of protective mucus and increase the functionality of local immune mechanisms, especially in respiratory and mucosal tissues.
In studies involving the combination of Houttuynia cordata with polysaccharides of Astragalus membranaceus and Ganoderma lucidum, an effect of modulation of innate immunity and enhancement of the proliferative activity of T-cells has been described. This interaction is classified as immunomodulatory and systemic, manifested in the support of the physiological activity of natural killers, activation of macrophages, and normalization of the level of pro-inflammatory mediators. The probable mechanism of synergy includes co-activation of Toll-like receptors and enhanced production of interleukins IL-12 and IFN-γ, which makes the combination particularly promising for immune regulation.
Some publications demonstrate that Houttuynia cordata in combination with quercetin derivatives, resveratrol, and extracts of Camellia sinensis enhances antioxidant protection at the tissue level through synergistic increases in the activity of SOD, CAT, and GPx enzymes. The combinations have a tissue-specific effect, manifesting particularly in models of oxidative stress in the skin and mucous membranes. The effect is mediated by activation of the antioxidant response through the Nrf2 pathway and a decrease in the expression of caspases responsible for apoptosis of cells damaged by free radicals.
A potentiating interaction of Houttuynia cordata with curcumin has also been recorded, due to the combined inhibition of PI3K/Akt and JAK/STAT pathways, which reduces the expression of pro-inflammatory proteins and enhances the apoptosis of inflammatory-transformed cells. This effect is systemic and cell-specific in nature and has been confirmed in in vitro studies on immunocompetent cells.

Geography of Use and Traditional Medicine – Houttuynia cordata
Houttuynia cordata is traditionally used in the ethnomedical and cultural practices of East and Southeast Asia, including the territories of China, Japan, Korea, Vietnam, Thailand, Laos, and northeastern India. In traditional Chinese medicine, the plant is known as Yu Xing Cao and is included in classical medicinal formulas as a "herbal antidote." Mentions of the use of Houttuynia are found in the pharmacopoeia of the Ming Dynasty (16th century), where the plant is described as cooling and cleansing. In Korea, the plant is called Chongnip and is used in the form of infusion, as well as a poultice and juice from fresh leaves. In Japanese folk medicine, it is known as Dokudami and is used as a cleansing, anti-suppurative, and restorative remedy.
In Vietnam, Houttuynia is considered a daily phytocomponent in the folk medicine of mountain ethnic groups, such as the Hmong and Thai. The leaves are used fresh, as well as in infusions and decoctions.
In the northern provinces of India (the states of Arunachal Pradesh, Assam, and Meghalaya), the plant is used in the practices of the Naga, Mishmi, and Apatani peoples, where it is an ingredient in traditional green pastes, wraps, and steam baths. In Laos and Thailand, Houttuynia leaves are also included in medicinal balms, oil extracts, and steam inhalations. In Thai folk medicine (Yai and Lanna traditions), the plant is known under the name "phak khao thong" and is used for body cleansing, cooling of internal energy, and restoration of dosha balance. In some rural communities, the leaves are used in smoking mixtures with herbs and in rituals of purifying the dwelling from diseased energy.
Historically, the plant was cultivated near monasteries, medicinal gardens, and household plots, where it was used for both medicinal and culinary purposes. Written mentions of Houttuynia as a medicinal remedy can be found in the Japanese treatise "Honzo Wamyo" of the Heian period (10th century), as well as in Vietnamese rural folklore records preserved in oral tradition. Among the indigenous peoples of Southern China (Miao, Yao), the plant was considered a talisman capable of purifying the space from diseases and "stale air," and was used in fumigation of dwellings during epidemics. In the shamanic rituals of the North Vietnamese peoples, Houttuynia leaves were hung at the entrance of the house to repel the "evil spirit."
In modern ethnomedical practices of the peoples of Thailand and Myanmar, the plant retains its status as a cleansing agent, used after childbirth, in body wraps with balm pastes, as well as in the form of steam sitz baths. In village culture, it is also used ritually — for purifying the space during days of eclipses and in periods when a person encounters "spiritual impurity." In the Lanna tradition, it is included in steam baths (saunas) as part of multi-herbal collections for women's and general health improvement.
Thus, Houttuynia cordata has a wide geography of traditional use, is deeply rooted in the cultural and medical practices of various ethnic groups of Asia, and combines medicinal, preventive, and symbolic significance in folk traditions.

Due to liver hepatosis, I developed a large red wart on my finger. Many deep pimples appeared on my face. I gained a lot of weight. I suffered from insomnia. Chronic fatigue did not go away. I was constantly taking a vitamin complex from Blackmores, without which I could not exist. As diverse as diseases are, there are also many ways to treat them. I was fortunate to start treatment with Vladimir. It lasted for 3 months. I was prescribed the following preparations: Reishi mushroom spores, Plukao (Houttuynia cordata extract), Krachaidam (Kaempferia parviflora), and Thunbergia laurifolia.
After starting the treatment, my sleep was immediately restored. Previously, I could not fall asleep even with sleeping pills. After a week, I felt a surge of energy (by that time, I had stopped taking the Blackmores vitamin complex). Gradually, I began to lose weight. The pimples on my face disappeared after 2 weeks. My red wart on my hand began to slowly shrink. But what came out of it shocked me. I have never seen anything like it. It looked like a tube of toothpaste, and this "paste" kept coming out, drying up, falling off, and immediately new one appearing. As of today, only a small dry sore remains from this wart. I sleep 8-9 hours now. My skin has become much better than it was before. I have lost a lot of weight. My hair has stopped falling out — before, it was shedding constantly. My energy has increased. I am very grateful to Vladimir for the correct treatment and for the useful advice, thanks to which I began to feel excellent. Volodya, may the love of the Universe adorn your life for many happy years.
