Standard Dosage (Powder): 1 gram of powder 3 times a day 30 minutes before meals, in a course of 30 days.

Enhanced Dosage (Powder): 2 grams of powder 3 times a day for malignant neoplasms of the breast, prostate, and hepatocellular carcinoma, in a course of up to 60 days under specialist supervision.

Residual high-molecular-weight fractions can penetrate the lymphatic system through M-cells of Peyer's patches, activating the mucosal immune system.

After entering the systemic circulation, the active fractions of polysaccharides predominantly bind to cells of the immune system, rather than circulating in free form. Metabolism of β-glucans occurs primarily in the liver, followed by excretion of metabolites with bile and partially with urine. Their binding activates intracellular signaling cascades, such as NF-κB, MAPK (ERK1/2, JNK, p38), which leads to modulation of the transcription of pro-inflammatory and anti-inflammatory cytokines, including IL-6, TNF-α, IL-1β, and IL-10. At the same time, stimulation of the expression of antioxidant defense enzymes is noted: superoxide dismutase (SOD), catalase, and glutathione peroxidase. This allows the fungus to be considered a systemic modulator of the immune response with the potential for restoring cellular homeostasis.

In neuroglial models, Hericium coralloides extracts demonstrate the ability to reduce the level of reactive oxygen species and lipoperoxidation, stabilize mitochondrial potential, and limit nitric oxide production by inhibiting inducible NO-synthase (iNOS). A decrease in the expression of the pro-inflammatory enzyme COX-2 and prostaglandin production is also observed, which confirms the anti-inflammatory direction of action through inhibition of key inflammation mediators. Neuroprotective action is realized through stimulation of brain-derived neurotrophic factor (BDNF) production, protection of neurons from apoptosis, and restoration of synaptic activity. Proposed targets are glutamate receptors, γ-aminobutyric acid receptors, as well as activation of the CREB/ERK signaling pathway in brain structures. The ability to inhibit acetylcholinesterase (AChE) activity has also been confirmed, which may contribute to maintaining acetylcholine levels in synapses.

At the level of the vascular endothelium and skin epithelium, Hericium coralloides is able to reduce the expression of adhesion molecules (VCAM-1, ICAM-1), inhibit the secretion of metalloproteinases (MMP-2 and MMP-9), prevent degradation of collagen structures, and stimulate the production of epidermal growth factor (EGF). These properties are mediated through JAK/STAT- and PI3K/Akt-dependent pathways and confirm the tissue-specific regenerative and reparative action of the fungus.

Finally, in vitro and in vivo data indicate a direct antiproliferative effect of individual fractions of Hericium coralloides extract due to induction of the G0/G1 cell cycle and activation of caspase-dependent apoptosis. Targets in this case are cytochrome c, caspase-3, Bax, and Bcl-2. This confirms the cascade mechanism of action at the cellular level with potential targeting of transformed cell growth.

Synergy — Hericium coralloides

Scientific data demonstrate a pronounced pharmacological synergy of Hericium coralloides with various natural substances in modulating immune, antioxidant, and neurotrophic activity. In in vitro experiments, it was established that combined use of extracts of Hericium coralloides and Ganoderma lucidum leads to potentiated release of IL-12 and IFN-γ, which indicates a synergistic enhancement of the Th1 response through joint activation of TLR- and Dectin-dependent pathways. This interaction is characterized by an additive and potentiating effect on cells of innate and adaptive immunity.

In combination with extracts of Hericium erinaceus, a modulating effect on neurotrophin production and glial growth factor expression is observed. This effect is mediated by synchronous activation of CREB and ERK1/2, as well as a decrease in the expression of pro-inflammatory mediators in microglia. Cellular-level synergy in astrocytes and neurons is suggested.

The combination of Hericium coralloides with green tea (Camellia sinensis) enhances the antioxidant effect through cumulative inhibition of lipid peroxidation and regulation of glutathione reductase activity. At the systemic level, tissue-specific protection is manifested, especially pronounced in epithelial structures of the skin and gastrointestinal tract.

Combined use with Curcuma longa demonstrates potentiation of anti-inflammatory activity through co-inhibition of NF-κB and iNOS, accompanied by a decrease in TNF-α and PGE2 production in monocytic cells. This effect has a systemic direction with a pronounced anti-inflammatory component. Also, data on additive activity have been recorded when combining Hericium coralloides with zinc, vitamin D, and selenium in relation to the regulation of immune response gene expression, which may be significant in multicomponent nutritional support regimens.

Geography of Use and Traditional Medicine — Hericium coralloides

Hericium coralloides has traditionally been used in a number of East Asian countries, including Korea, China, Japan, and Mongolia. References to its use are also recorded in ethnographic materials from Eastern Europe and Siberia. In Korea, the mushroom is known as "Neungi" and is highly valued in traditional medicine as a nutritious product and strengthening agent. In Chinese folk medicine, it is classified as a tonic substance and is used primarily in decoction form, as well as in soups and infusions. Japanese sources record its consumption in cooked form as part of health dishes aimed at maintaining the general condition of the body and longevity.

In North Asian regions of Russia, including Altai and Southern Siberia, the mushroom was traditionally collected by the Khanty, Buryat, and Tuvan peoples. It was considered a "pure mushroom," suitable not only for food but also as a component of steam procedures and water douches in the bathhouse. In Slavic village practices, Hericium coralloides was used in alcoholic tinctures and consumed in the form of dried slices ground into powder and brewed in clay dishes. Among the Old Believers, there was a belief that this mushroom protects against "internal exhaustion" and cleanses the "clouded mind."

In some cultures of Eastern Europe, the mushroom was included in ritual dishes for harvest festivals and was considered a symbol of natural strength and health. Archaeoethnobotanical finds from southern Poland and Western Ukraine indicate possible consumption of Sparassis as early as the 15th–16th centuries, although written sources on its targeted medical use appear only in the 19th century in herbal books of the Russian Empire. In them, it is described as "forest cabbage," strengthening the body in the cold season.

In Mongolian tradition, the mushroom is less common; however, some ethnographic sources record its inclusion in dry mixtures for winter storage and as a component in decoctions consumed after exhausting trips or physical overload. In certain ritual practices, the mushroom was used as "cleansing" food during transitional rites — primarily in the South Siberian shamanic environment.

In a non-medical and esoteric context, Hericium coralloides was used as a protective amulet in some regions. Its dried fruiting bodies were hung above the entrance to the house or placed at the head of a sick person for "restoring clarity of mind." In some villages of Poland and Belarus, references have been preserved to the use of the mushroom in ritual fumigations, when it was burned in a stove or on coals during the purification of premises from "evil breath" and "heavy memory." In Siberian shamanic rituals, the mushroom was part of fragrant offerings presented to nature spirits.
