Toxicity and Biosafety — Tatarinow's Sweet Flag (Acorus tatarinowii)

Biosafety studies of Acorus tatarinowii have shown that the aqueous and alcoholic extracts of the plant have low acute toxicity upon oral administration. In an experiment on laboratory animals (mice, rats), the median lethal dose (LD₅₀) of the aqueous extract of Acorus tatarinowii upon oral administration exceeded 18 g/kg body weight, which corresponds to the classification of substances with very low toxicity. For the alcohol extract, the LD₅₀ values were also in the safe range, exceeding 12 g/kg body weight.

Additional chronic toxicity studies showed the absence of pronounced hepato- and nephrotoxic effects. No signs of tissue degeneration, inflammatory reactions or changes in blood formula were detected in histological samples. The absence of mutagenicity and cytotoxic effects on liver and brain cell cultures at subtoxic concentrations was also confirmed.

Thus, Acorus tatarinowii can be classified as a substance with a high biosafety profile when the recommended therapeutic dosages are observed.

Pharmacodynamics — Tatarinow's Sweet Flag (Acorus tatarinowii)

Tatarinow's sweet flag (Acorus tatarinowii) demonstrates a wide range of pharmacodynamic activity, confirmed by experimental and biochemical studies. The basis of action is essential oils and phenylpropanoid compounds, mainly α-asarone, β-asarone and other monoterpenoids, as well as flavonoids and alkaloid-like compounds. Their activity covers central and peripheral regulatory mechanisms, including interaction with key enzyme systems and receptor structures.

At the level of the central nervous system, Acorus tatarinowii has a pronounced modulating effect on cholinergic neurotransmission by inhibiting acetylcholinesterase activity. This leads to an increase in the concentration of acetylcholine in the synaptic cleft and activation of muscarinic and nicotinic receptors, which determines the overall nootropic, antiamnestic and modulating effect. In addition, weak antagonistic effects on NMDA receptors have been identified, which apparently reduces excessive neurotransmitter load and protects neurons from excitotoxicity.

At the biochemical level, plant extracts regulate oxidative stress levels, increasing the activity of superoxide dismutase (SOD), catalase and glutathione peroxidase, while reducing the concentration of malondialdehyde and other lipid peroxidation products.
